Micropropagation in Phaius Tankervilliae Orchids

Abstract/ Executive Summary:

The present study was conducted to investigate the effect of IAA and in combination with IBA on the growth and development of Phaius tankervilliae orchid. Data on shoot length,leaf number,root number and root length was recorded. The highest shoot length, leaf number and root length were obtained from the treatment, 1.0 mg/l IAA + 1mg/l IBA + 0.1% charcoal. Root number was found statistically higher when media were supplemented with charcoal.The activated charcoal enhanced root formation. For ex vitro establishment, maximum survival percentage was recorded when plantlets were hardened off for 7 days at room temperature

Objectives:

To develop an efficient protocol for mass propagation of this important local orchid Phaius

Methodology:

Selfed pods were collected from mature plants and were washed with running tap water. They were then surface sterilized by submerging them in 0.1% HgCl₂ solution with 2-3 drops of Tween -20 for 15 min.The pods were dipped in absolute ethanol and flamed for a few seconds holding with forceps. They were then splitted longitudinally with a sterile scalpel and the seeds were taken by forceps and transferred to the agar silidified MS medium.Different concentrations of IAA,IBA and BAP were used for this experiment

Highest shoot length, leaf number and root length were obtained from the treatment, 1.0 mg/l IAA, 1.0 mg/l IBA and 0.1% charcoal.Number of root was found statistically high when medium was supplemented with charcoal.Plantlets survival percentage was increased with the increase in the days of hardening.Maximum survival percentage(93%)was recorded when plantlets were hardened.
