In Vitro Conservation of Mint (2015--2016)

Abstract/ Executive Summary:

The aim of the study was to increase the in vitro conservation period of mint (Mentha spp.) at 22 ± 1°C. Nodal explants of three genotypes viz. MP-1, MP-2 and MP-3 of mint were cultured on MS medium supplemented with 20 mg/l sorbitol. All the genotypes produced 100% roots within 7-9 days and shoots within 4 to 8 days. The maximum number of leaflets (89.6), shoots (8.6) and roots was observed in MP-2 and the minimum number of leaflets (47.2) and roots (3.4) was found in MP-1. The genotypes produced 5.1 to 10.6 cm shoot length with an average of 7.2 cm. The genotypes produced an average of 63 leaflets and 5.8 shoots. MP-1 performed better in minimal growth of in vitro conservation. This in vitro conservation technique can be used for conservation of mint.

Objectives:

An attempt has been made to increase the in vitro conservation period of mint genotypes.

Methodology:

The experiment was conducted with three genotypes viz. MP-1, MP-2 and MP-3 of mint at the In vitro Conservation Laboratory of Plant Genetic Resources Centre, BARI, Joydebpur Gazipur. The genotypes have been maintaining at the Laboratory for three years. Basal media contained MS macro and microelements including vitamins (Murashige and Skoog 1962). Four weeks aged plantlets were taken on sterilized petridish. Nodal segments with two leaflets were cut aseptically under laminar flow and these segments were used as explants. MS basal salts 44.0519 mg/l, 20 mg/l sorbitol, and 30 g sucrose were dissolved in distilled water and mixed well. The pH of the medium was adjusted to 5.8 before autoclaving and the media were solidified with agar @ 8 g/l. Fifteen ml of media was poured into each culture tube (25 x 150 mm). The medium was autoclaved at 1.06 kg/cm² pressure and 121°C for 20 min. One nodal explant was placed vertically on the medium keeping the position of the tip upsides and gently pressed into the surface of the sterilized culture medium. The nodal explants were incubated for three months in a culture room at 22 ± 1°C under about 2000 lux light provided by cool-white fluorescent tubes maintaining a 16/8 hr alternate light/dark cycle and 60-70% relative humidity. The culture tubes were checked daily to note the morphogenic development and the data were recorded on 8 parameters. The experimental setup was in a completely randomized design with five replications and each replication consisted of 5 explants. Data were subjected to ANOVA using the General Linear Model procedure (Gomez and Gomez, 1984). Mean separations were carried out by LSD at 1% using the MSTAT program.

Achievement/Findings:

The highest percentage (100%) of shoot and root initiation (100%) was observed in all MP-1, MP-2, and MP-3. Days to root initiation were ranged from 7 (MP-2) to 9 days (MP-1). After five months of culture in MS containing 20mg/l sorbitol, MP-2 produced the maximum leaflets (89.6), and the minimum leaflets were obtained from MP-1 (47.2). The genotypes produced 3.8 to 8.6 shoots and 4.8 to 10.4 roots/plantlet. The maximum shoot length was exhibited in MP-2 (10.6 cm) and the minimum from MP-1 (5.1 cm) (Table1). Among the genotypes, MP-1 performed better in minimal growth for shoot proliferation through node culture. Sorbitol (20 g/l) with 3% sucrose showed good performance in vitro conservation of mint genotypes. MP-1 performed better than the remaining genotypes for minimal growth in mint through node culture. Sorbitol can be used in vitro conservation of mint.
