Development of an Efficient Regeneration System of Banana (2016-2017)

Abstract/ Executive Summary:

The present study was undertaken with a view to establish a reliable regeneration protocol of banana (Musa spp.). Bottom portion and whole male flower buds were used as explant for callus induction. Explants were cultured on MS medium supplemented with different concentrations of 2, 4-D singly and in combination with different concentrations of IAA and NAA. In Bottom portion of male flower buds, among the four different treatments, the highest percentage of explant produced callus in T₂ treatment (56%) compared to other treatments. In whole male flower buds, the highest percentage of explant produced callus in T₁ and T₂ treatment (100%) compared to other treatment.

Objectives:

To develop an efficient regeneration protocol for Bananas which will be used for the development of banana transgenic with desirable traits.

Methodology:

Male flower buds of banana variety Maher Sagar were used as source material. The fresh male flower buds were collected at the time of complete bunch emergence. Outer enveloping bracts were removed and surfaces were cleaned with absolute alcohol. Under laminar air flow cabinet, the remaining outer enveloping bracts were removed and the inner part (2-3 cm in length) containing male flower primordia were isolated. Individual male flower primordia were excised and cultured on MS medium supplemented with different concentrations of 2, 4-D and in combination with IAA and NAA for initiation of callus as well as regeneration of shoot. The p^H of all media was adjusted to 5.8 before autoclaving. The media were autoclaved at 121⁰C and 1.06 kg cm^-2 pressure for 20 minutes. All cultures were incubated at 25±2⁰C in complete darkness and maintained on the same medium without subculture until callus formed.

Bottom portion of individual male flower buds were cultured on MS medium supplemented with four different concentrations of 2, 4-D showed enlargement of the explants and the development of whitish callus after one month of inoculation. Among the treatments, maximum callus was found in T₂ treatment (56%) followed by T₄ (50%). The lowest calli were noticed in treatment T₁ (31%). Individual male flower bud were cultured on MS Medium supplemented with 2, 4-D and in combination with IAA and NAA to find out the suitable culture medium for callus induction. Male flower buds cultured on MS medium supplemented with 2, 4-D and four different concentrations of IAA and NAA showed enlargement of the floral primordia and the development of whitish callus after one month of inoculation. Among the treatments, maximum callus was found in T₁ and T₂ treatment (100%). The lowest calli were noticed in treatment T₃ (44%). The calli were then separated and transferred to fresh MS medium containing different concentrations of BAP, IAA and casein hydrolysate in order to find suitable media composition for regeneration of shoots. In this medium, all calli turns to greenish within two weeks of culture. But no shoots were produced.
