Collection and Assessment of Arbuscular Mycorrhizal Fungi in Different Field Crops (2017-2018)

Abstract/ Executive Summary:

Rhizosphere soils from 24 field crops were collected from Central Agricultural Research Station, BARI, Gazipur and were assessed during 2017-2018 for determining root colonization (%) and spore population (100 g^-1 soil) of AM fungi. Kalozira showed the the highest number of spores (94.7 100 g^-1 soil) and Barley gave the highest root colonization (30.0%).

Objectives:

To make a general survey of diversity in mycorrhizal association with agricultural crops and spore population in rhizosphere soils in different Agro Ecological Zones of Bangladesh.
To understand the dynamics of AM fungal root colonization and spore population for preparing a suitable arbuscular mycorrhizal inoculum for different crops.

Methodology:

Roots and rhizosphere soil samples were collected from BARI Central Farm, Gazipur during rabi 2017-2018 when abundant plants were available in the fields. Five plants for each species were sampled. Plant roots were dug out, washed thoroughly with water to remove the adhering soil particles and then cut into 1 cm long segments. The root samples were then preserved in screw cap test tubes with 50% ethanol for future use. Soil samples were collected up to a depth of 0-15 cm with roots (3 samples/crop). Then the collected samples were air-dried, packed in airtight polyethylene bags and stored at 4^°C for assessing spore density. The percentage of AM infection was estimated by root slide technique (Read et al., 1976). Assessment of spore population was done by following the Wet Sieving and Decanting Method (Gerdemann and Nicolson, 1963).

Achievement/Findings:

Different crop species showed different percentages of root colonization by AM fungi. Percent root colonization in different agricultural crops varied from 20.00-30.00%. More than 20.00% root colonization was recorded with Barley, Bhutta and Gom. Other crops showed 20% root colonization. The colonization rate of AM fungi is different with the plant species due to differences in the structure of root system (Hetrick et al. (1992). More crops covering different AEZs should be included in next year study. This programme will be continued in the next year.
