Cytological Study of Peripheral Blood of Freshwater Mud Eel, Monopterus Cuchia (hamilton)

Abstract/ Executive Summary:

Various types of haematocytes were observed in the peripheral blood of freshwater air breathing mud eel, Monopterus cuchia. They have been identified and characterized using morphological, morphometric and cytochemical techniques, for the first time in Bangladesh. The mature erythrocytes were oval to round shaped with centrally placed deeply stained nucleus. Lymphocytes were spherical having large, eccentric and round nucleus. Lymphocytes were divided into two groups, large and small lymphocytes. The monocytes were rounded pale grayish-blue coloured with eccentric and auricula purple coloured nuclei. The tiny structured thrombocytes were oval and convex in shape with condensed nuclei and with very little cytoplasm. The polymorphonuclear leucocytes were neutrophils that were large and round in shape with fine granules in cytoplasm having eccentric nuclei. The shape of the nuclei varied from spherical, oval, bi-lobed or tri-lobed. The cytoplasm stained whitish, white- gray or bluish-gray colour and nucleus stained purple colour with Wright and Giemsa.

Objectives:

To identification of different types of cell found in peripheral blood of the fish with their possible classification.

Methodology:

Apparently healthy freshwater mud eel, Monopterus cuchia was chosen to study haematocytes. They were stocked in the cemented cisterns of the Faculty of Fisheries, Bangladesh Agricultural University, Mymensingh, Bangladesh. Thirty live fish were sampled taking ten fish randomly in each month during April to June 2002. Average length and weight of the fish were recorded. In order to minimize an error in normal blood values, fish were caught gently in small scoop net avoiding stress and transferred into a plastic bucket containing the same water where they were stocked. For anesthetizing the fish, a mixture of clove oil: ethanol: water was used in the ratio of 2:18:9980 (0.02%) as reported. At first 0.2 ml of clove oil was mixed with 1.8 ml of ethanol. This was then mixed with distilled water to make the volume one liter. An individual fish was released in this anaesthetic to anaesthetize it within 10 min. After anesthetizing the fish, blood was collected from the caudal vein with a sterile disposable plastic syringe coated with 3.6 % sodium citrate as an anticoagulant. To avoid contamination with mucus and water the area of insertion of needle of syringe was wiped with cotton soaked in 70% alcohol before and after the collection of blood. This blood sample was used for blood film preparation and cell count. Thin blood films were prepared immediately after collection on previously cleaned glass slides in the following way: a drop of blood was placed on one side of the slide. Another slide was placed in front of the drop in an inclined vertical position making 45° angle towards the drop in such a way that the slide just touches the drop. This slide was then pushed to the other side of the flat slide to prepare a smear. All microscopic studies like measurement and characterization of cells and differential cell counts were done with such stained slides under a compound microscope at different magnifications as and where needed. An occulomicrometer was used for measuring the cells. A total of 100 leucocytes were counted under microscope at (100x) for each sample and the percentage of different types of leucocytes was calculated. These slides were used for the photograph of different blood cells.

Achievement/Findings:

Like other animals, fish haematocytes are index of fish health. For example, in case of infections, the numbers of lymphocytes, monocytes and macrophages increase. Also they play an important role in the defense mechanism of fish especially neutrophil and monocyte like cells show phagocytic activity against foreign pathogenic agent. Thus the findings of present study will help fish pathologists to determine the health status of Monopterus cuchia.
