Comparative Efficacy of Experimentally Prepared V4hrnd Vaccine with Commercial V4hr Newcastle Disease Vaccine and Bcrdv Vaccine

Abstract/ Executive Summary:

A serological immune response and protection in broiler birds against Newcastle disease with experimental V₄HRND vaccine was compared with commercial V,HR Newcastle disease vaccine and locally produced conventionallentogenic F- strain Baby Chick Ranikhet Disease vaccine (BCRDV, DLS, Dhaka) of Bangladesh. A total 40 broiler birds were divided into four groups (W, X, Y and Z) and each group consisting of 10 birds. Each bird of group W, X and Y was vaccinated with experimental V₄HRND, commercial V₄HR and BCRDV vaccines, respectively twice at 7 and 22 days of age while group Z was kept as control. Sera samples were collected from all vaccinated and control birds at 4,7,14, 17, 21, 29, 32, and 37days of age. Antibody titers were determined by haemagglutination inhibition (HI) and enzyme linked immunosorbant assay (ELISA) tests. From this study it was found that BCRDV, experimental V₄HRND and commercial V₄HR produced 7.25±.95, 5.75±1.25, 5.25±0.96 HI antibody titers and the ELISA titers were 3.16±0.03, 2.85±0.10 and 2.82±0.03 respectively. BCRDV, experimental V₄HRND and commercial V₄HR gave 100%,80% and 60% protection.

Objectives:

To compare the efficacy of serological responses and protection among the locally produced conventional F strain (BCRDV) vaccine, commercial and experimental V₄HRND vaccine in Broiler chicks.

Methodology:

Forty day-old Broiler chicks with the history of vaccination of parent stock against Newcastle disease (ND) were purchased from Kazi Farms Ltd. Dhaka and reared at the experimental sheds in the Department of Microbiology and Hygiene, BAU, Mymensingh during the period from November 2003, to March 2004. Total birds were divided into four groups (W, X, Y and Z) consisting of 10 birds in each group. The Australian heat resistant V. strain of ND virus (V₄HRNDV) and commercial vaccine were obtained with the courtesy of small holder Livestock Development Project-2 (SLDP- 2) and F-strain (Baby Chick Ranikhet Disease Vaccine BCRDV) vaccine was obtained from the Directorate of Livestock Services (DLS). Each bird of group W, X and Y was vaccinated intraocularly @ 0.1ml/bird with experimental V₄HRND, commercial V₄HR and BCRDV vaccines respectively at the age of 7 and 22 days of age and group Z was kept as unvaccinated control. Sera samples were collected from each bird of all groups at 4, 7, 14, 17, 21, 29, 32, and 37days of age. Antibody titers of the sera samples of the vaccinated and control group of birds were determined by haemagglutination inhibition (HI) and enzyme linked immunosorbant assay (ELISA) tests. Each bird of vaccinated and nonvaccinated group was challenged with virulent field isolate of NDV obtained from the Department of Microbiology and Hygiene, BAU, Mymensingh, after 2 weeks of secondary vaccination @ 0.1 ml of 10⁵ ELD₅₀ intraocularly. The challenged birds were kept in the isolated room and observed for 7 days twice daily. Any bird showing the symptoms of ND was recorded and postmortem lesions of ND were examined in all of the dead birds. Results were analyzed statistically by using Computerized Statistical Program (SPSS).

Achievement/Findings:

From the study it may be concluded that although BCROV is more effective than experimental V₄HRNO and commercial V,HR vaccines, but it needs maintenance of cold chain during transportation and use, which is not always possible in our country. On the contrary V₄HRNO vaccine could be used in our country to protect NO without maintaining strict cold chain system, because it is also immunogenic, gives satisfactory level of protection, easy to transport and there is no possibility to produce any disease. It is difficult to eradicate the Newcastle disease from Bangladesh, as such to protect ND routine vaccination program should be strictly followed and effective vaccination with V₄HRNO vaccine might be a better choice.
