In Vitro Clonal Propagation of Jackfruit

Abstract/ Executive Summary:

Clonal propagation technique of Artocarpus heterophyllus Lam. through in vitro culture has been developed using apical bud from mature tree. MS basal medium supplemented with 5.0 mM BAP was found better tor multiplication and elongation (If shoot. The maximum rooting was observed on half MS medium supplemented with 10 mM IBA. Seventy two per cent plantlets survived when the regenerated plantlets were transfered to the soil.

Objectives:

To establish an in vitro rapid propagation protocol of jackfruit using apical bud from elite mature tree.

Methodology:

Apical bud explants were collected from actively growing branches of 10-12 years old elite fruiting tree grown at the fruit farm of Bangladesh Agricultural Research Institute, Joydebpur. Gazipur. Unopened buds were collected and washed thoroughly under running tap water for 30 minutes. After that, the buds were treated with 30% sodium hypochlorite solution for 15 minutes and rinsed 4 to 5 times with sterile distilled water. Finally. the explants were prepared under clean bench by removing the outer covering of green stipules and excising the inner bud enclosed within the creamy white stipules. The prepared explants were' cultured on MS (Murashigc and Skoog, 1962) medium supplemented with different concentrations of BAP (0. 1.0. 2.5. 5.0, 7.5 and 10.0 mM) for shoot proliferation and multiplication. Sucrose (3%) and Agar (0.8%) were also added to the media. Half MS medium and different concentrations of auxins (0. 5.0. 7.5 and 10.0 mM of IBA and N:A separately) were used for adventitious root formation. The pH of all media was adjusted to 5.8. For shoot multiplication and rooting, the media were autoclaved at 121 DC and 1.2 kg/cm² pressure for 25 minutes. The cultures were grown at 25DC (± 1⁰C) under 16h photoperiod using white fluorescent tubes. Each treatment of shoot proliferation and rooting consisted of 20 replicates. All the cultures were regularly subcultured on to the same fresh medium at 4 weeks interval and individual shoots were placed in the rooting medium. After rooting the plantlets were transferred to glass tubes containing sterile sand, soil and vermiculite (1: 1: 1). The glass tube was covered with lid and kept in growth chamber for producing profuge functional roots. Half strength MS solution was sprayed into the tube as and when required. After 45 days, hardening was done for one week keeping the tubes in the same growth chamber by opening the lid slightly. The plantlets were then transferred to small plastic pots containing sterile sand, soil and cowdung (1: 1: 1) and covered with polyethylene paper to maintain high humidity for 15 days in laboratory at room temperature. Two months after the plants were transferred to the bigger plastic pots (12.5 x 17.5 cm) containing coarse sand, soil and well decomposed cowdung in 1: 1: 1 ratio and kept in net house for their growth. Watering was done as and when required.
