Insecticidal Activity of Ipomoea Maxima L. Extracts Against the Pulse Beetle, Callosobruchus Maculatus F.

Abstract/ Executive Summary:

Leaf, stem and root of Ipomoea maxima L. were extracted in four different organic solvents (petroleum spirit, ethyl acetate, acetone and methanol). The contact toxicity of extracts were tested and found that it was very effective in killing adult Callosobruchus maculatus F. In all the cases the order of toxicity were petroleum spirit> ethyl acetate> acetone> methanol. Males were found more susceptible than female. Leaf and stem extracts were more effective in comparison to root extracts.

Objectives:

To evaluate the activity of extracts of I. maxima against the adult C. maculatus.

Methodology:

The collected plant parts were dried in an oven and made into fine dust in a hand-grinding machine. The dust of plant parts were extracted in four different organic solvents viz. petroleum spirit (pt. spt), ethyl acetate (EtOAc), acetone and methanol (MeOH). The extracts were dried in a vacuum rotary evaporator at 40°C under reduced pressure. Four different doses i.e., 0.01875, 0.03750, 0.07500 and 0.15000 g for pt. spt. and EtoAc; 0.025, 0.050, 0.10 and 0.20 g for acetone and MeOH extracts were weighted by an electric balance and dissolved in requisite amount of respective solvents. The doses were transferred to µg/cm² by measuring the dry-weight of extracted materials and divided by the surface area of petridish (9 cm diameter). The test insects were exposed to the residual film technique. Extract was dispersed into each petridish for each dose. After evaporating the solvent from the petridish 30 same aged (0-24 hours old) adult male and female C. maculatus were released into each petridish. Control lines for male and female were maintained with respective solvents only. The experiments were replicated three times by same procedure at 30±1°C and 70% relative humidity. Mortality was recorded after 24, 48 and 72 hours exposure. The beetles were reared in Black gram (Phaseolus mungo L.) seeds in Petri dish (15 cm diameter) at 30±1°C and 70% relative humidity (rh).

Achievement/Findings:

Laboratory experiments with I. maxima showed that this plant has got insecticidal activity against the pulse beetle C. maculatus. The results of the present research indicate the possible use of I. maxima leaf, stem and root extracts to control C. maculatus in storage.
