Study of Biotypes of Macrophomina Phaseolina in Controlled Condition

Abstract/ Executive Summary:

The experiment was conducted in Green house, Central station, Dhaka to find out the sources for resistant materials against Macrophomina phaseolina. during 2006 jute seasons. Nine C. capsularis, 8 C. olitorius and 7 kenaf and mseta lines/varieties were screened in green house against virulent races (biotypes) of M. phaseolina. Mesta variety HS-24 advanced kenaf lines KE-3 and C. olitorius line SDL-3 and one accession 4620 had tolerant reaction against two biotype MS-6 and MS-9 of M. phaseolina.

Objectives:

1.To find out the sources for resistant materials against Macrophomina phaseolina and

2.To help the breeders to develop the resistant varieties against Macrophomina phaseolina.

Methodology:

A total of 25 accessions of Corchorus sp. kenaf and mesta was sown in the pots following CRD with three replications. Three plants (30-40 days old) of each treatment was inoculated on 3 spots of each plant with 48 hours old culture of each race grown in PDA medium. Circular block ( 2mm diameter) was cut with a block cutter. One block was placed on each spot on stem after pricking with a needle. The pots were kept inside the humid chamber in the green house for 48hours where 90% humidity ( about 90% & temperature 30^oC was maintained. After 48 hours pots were taken out of the chamber. Records were kept: a)Necrotic lesions was measured in mm after 7-10 days of inoculation. b)Disease reaction was assessed on the basis of lesion size & grouped as resistant (R), moderately resistant (MR ), Susceptible (S ) and Highly Susceptible (HS ). c)Seeds from resistant and moderately resistant treatments was collected.
